PCR tubes/plates are commonly available in white and transparent, and in traditional PCR experiments, there is not much difference in the use of the two, but in quantitative PCR experiments, the sensitivity and consistency of white tubes are relatively higher for qPCR instruments that require apical signal reading. Let's briefly explain why.
The core data of qPCR is the Ct value, and the fluorescence signal is the data source for the Ct value calculation, so it is critical that the fluorescence in the tube is well transmitted to the detector. The white tube wall prevents fluorescence from passing through the tube wall and refraction, thereby preventing fluorescence from transmitting to the heating module, that is, the metal hole wall where the tube is placed, and the transparent tube will cause fluorescence to be absorbed by the module or inconsistent reflection. Therefore, compared with transparent tubes, the use of white tubes can effectively avoid signal interference, so as to collect stronger and more focused fluorescence signals.

Unskirted can be used universally on all models, while fully skirted PCR boards are typically used in automated workstations. Simply put, the significance of skirting is more to facilitate industrialized and automated batch processing, and the combination of robotic arms.
